Label-free detection and mass measurement of single biomolecules in solution has made dramatic advances over the past decades. I will discuss recent efforts aimed at understanding the fundamental nature of the interaction, rationalizing image formation and contrast in mass photometry all the way to the atomic structure of biomolecules. These results help understand both the magnitude and shape dependence of the observed signals. I will also present recent efforts for single molecule detection using optical holography and compare it with the status quo of common path interferometry. Taken together, these results quantify both the status quo and future opportunities in this growing field.
