Fluorescence-Detected Mid-Infrared Photothermal Microscopy
Fluorescence is a remarkably effective “thermometer” for photothermal microscopy. Changes of 1–2% in quantum efficiency per oC are routinely observed, yielding sensitivity to transient temperature changes of only a few degrees within a few ns over a few hundred nm spot size. Furthermore, fluorescence yields a diffraction-limited point-spread function with tight lateral and axial confinement in photothermal microscopy.
In this presentation, instrumentation and methods are presented for using fluorescence to transduce photothermal absorption to perform chemically selective microscopy. Particular emphasis is placed on integration of broad-band/multi-wavelength infrared sources for multiplexed hyperspectral imaging. Applications are presented for analysis of pharmaceutical materials and for biological imaging. Recent progress and critical assessment of figures of merit will be present.
